How does one edit /etc/cron.daily/tklbam-backup to define a time of day for the daily backup to run. I want the daily to start each day between 10 and 10:30pm.

How can I make sure the monthly full backups run on the weekend?

By default it will run a full backup once the last full backup is over 30 days old. I'm sure that you could make it work differently but I must admit that OTTOMH I'm not 100% sure...

The TKLBAM FAQ sort of answers some of your questions(s) but not quite exactly... Perhaps it's worth have a read over the TKLBAM docs?

Worst case, I'm sure that you could create your own cron job to launch it exactly when you want...
